Presumably XP, presumably Outlook Express?

If my presumptions are correct:
Go to Start > Run, type regedit and press Enter then navigate to registry key
HKEY_CURRENT_USER\Software\Microsoft\Windows\CurrentVersion\UnreadMail\[Your
E-Mail Address]

In the right pane, right-click MessageCount, click Modify and change the value to 0 (zero).

Or if anyone isn't confident about playing around in the registry, Tweakui click here will do it for you.

Open Tweakui, click on the "Repair" option, then choose "Repair unread mail count" from the dropdown menu and click the "Repair Now" button.
