missing dll, 'ugzyxd'

Recently on starting up I get the error message "unable to find C:\windows\system\ugzyxd.dll". When I click OK the PC runs normally.
Looking on the startup tab in MSCONFIG the file is there as ugzyxd.dll,Init1.

It may have something to do with a trojan that was recently spotted by AVG.

Is it ok to 'uncheck' this item?

I can't imagine any legitimate program having a .dll file as "ugzyxd." It might well have something to do with that trojan.

I've looked on click here and there isn't anything like it on there.

Yes it'll should be okay to untick the item in msconfig. The reason you're getting the error message on startup is because Windows is searching for the file. If you untick it in msconfig, Windows will no longer try to start it at bootup.
