Home page hijacked by safetyuptodate.net

I cannot access my Google.uk homepage, its beenhijacked by a fake page titled Internet Security which appears with a warning that System 32 is compromised with an ad. to download Malware Wipe or Pest Trap. In addition System 32 file page appears during start up sequence. Other malware including spywarequake, zlob download, adware spy sheriff seem to have been successfuly removed by Spybot, Norton and Adaware, the latter finding safteyuptodate.net which it seems cannot be removed. Windows defender cannot spot anything.
I have seen references to this being a type of Smitfraud family of hijackers and specialised software is available (but not officially verified) to remove this spyware but it seems complicated and none of the standard spyware packages can deal with it.
How should I remove this spyware, and is it now intrinsically harmful, that is, is my computer's system 32 security really compromised?
