Formatting hard drive

Wondering if anyone can shed any light here. I am trying to format my hard drive (its actually the E: drive, i have two hard drives, the other, bigger one, the master is the C:)at the command prompt, but after trying to format i get the message saying "format cannot run because the volume is in use by another process, format may run if this volume is dismounted first. All opened handles to this volume would then be invalid."
It then asks:-"Would i like to force a dismount on this volume Y/N>"
After clicking yes it says "cannot lock the drive, the volume is still in use". I have an seagate ST313021A with a gigabyte 8IR2003 motherboard and windows XP if that is any help.
Any help would be appreciated.

What command prompt? How are you reaching the prompt?

Isnt your E drive showing in My Computer if so just right click and format

I am accessing the command prompt through going to run and typing cmd.

Strangely when i right click the e:drive the format option is greyed out.
